Effect of Bax/Bcl-2mRNA Expression of Wendan Decoction Medicated Serum on PC12 Cells

Objective:To study the effect of Connexin Bax/Bcl-2mRNA expression of Wendan decoction medicated serum onPC12 cells.Methods:1、Medicated serum preparation: 30 SD rats were divided three groups by random.They were Wendan Decoction high-dosage group(A),Wendan Decoction middle-dosage group(B) and Wendan Decoction low-dosage group(C) respective ly.A 、 B and C groups were lavaged Wendan Decoction every day.After 8 days all rats were put to death.Then take blood from femoral artery,which centrifuge to take serum,and inactivate and sterilize them to set aside.2 、 PC12 cells group training : The PC12 cells will divided into 4 groups,they are comparison group(common culture media)、 WenDan decoction high-dosage medicated serum group 、 WenDan decoction middle-dosage medicated serum group and WenDan decoction low-dosage medicated serum group.Accord ing to the grouping of corresponding change medicated serum to train after 16 h,then extract total mRNA.Using drug-containing se rum to train PC12 cells after 16 hours,application RT-PCR half quantitative assay to test the expression of Bax mRNA and Bcl-2 mRNA by Wen Dan Decoction medicated serum for PC12 cells.Results : the Bax mRNA expression of Wendan decoction Groups was lower than that of comparison group(P 0.05 or P 0.01).And the Bcl-2 mRNA expression of Wendan Decoction high-dosagel group and Wendan Decoction middle-dosage group were rise compared with that of comparison group(P 0.05 or P 0.01).Conclu sions : Wendan Decoction can reduce the expression of Bax mRNA and rise the expression of Bcl-2mRNA on the PC12 cells.
