Comparison of different extraction methods of total DNA in Mangifera indica

Abstract

In order to research the effects of different extraction methods on extraction quality of total DNA in Mangifera indica,taking leaves in eleven cultivars of M.indica as materials,total DNA was extracted by the methods of SDS,CTAB and improved CTAB,and the concentration of total DNA,OD260/280,OD260/230,agarose gel electrophoresis test results,ISSR amplification products were compared and analyzed.The results indicate that the improved CTAB method can well remove the impurities in M.indica leaves,for example polyphenol,polysaccharide,tannin,pigment,and so on.The total extracted DNA concentration is up to 387 ng/μL,OD260/280 is from 1.81 to 1.87,and OD260/230 is from 2.01 to 2.09.The ISSR productions of total DNA extracted by SDS and CTAB method have fewer bands,but amplified bands of total DNA extracted by improved CTAB method is clear and has high polymorphism.In the three methods,the improved CTAB method has the best extraction effect,and it can be used for the subsequent researches.
