Abstract
The genomic DNA of Illicium verum were extracted with 4 plant genomic DNA extraction methods, namely CTAB,SDS,high concentration salt and reagent kit.The results showed that the improved CTAB could achieve promising extraction effect. The modification included using CTAB-free buffer to wash out for 1~2 times prior to the procedure of adding CTAB extraction buffer;adjusting the used content of CTAB in extraction buffer from 2 % to 3 %;adding the extraction buffer to 60 times of the dry weight of leaf sample from 10 times.Using the DNA extracted by the improved CTAB method, through RAPD-PCR amplification, clear bands were achieved;and the quality of extracted DNA could sufficiently meet the requirements of further molecular marker experiment.
