Comparative studies of mtDNAs from a D~(2)-type cytoplasmic male sterile wheat line, its maintainer and restorer lines

Abstract
msD 2-CA8057 is a recently developed D 2-type cytoplasmic male sterile (CMS) wheat line. Its cytoplasm was dervied from Ae. crassa 6x. Through RFLP and RAPD analyses, its mtDNA was compared with those of its maintainer line, CA8057, and restorer line, Bao-769-22-6, of which the cytoplasms are both from T. aestivum . The organization of the mtDNA from this CMS line was different from thoseof the two normal wheat lines. Southern hybridization result revealed polymorphisms among their atp A, atp 9, cob and cox Ⅱ genes, while the organizations of mtDNAs from the maintainer line and the restorer line were very similar. RAPD analysis further confirmed this result. This supports that the cytoplasmic genetic characters of this CMS line were derived from a wild type cytoplasm. Such cytoplasmic difference may be related to the fertility of this sterile line.
