Abstract
Objective To study whether peroxisome proliferator-activated receptor gamma(PPARγ)agonist rosiglitazone has protective effects on diabetic nephropathy and to explore its possible mechanism.Methods Ten SOD rats were regarded as control group.All rats with high-sucrose-high-fat diet were injected STZ(35 mg/kg)to induce diabetic rat model.Then diabetic rats were randomly assigned to 3 diabetic groups treated with a low dose and a high dose RGZ,and without RGZ.At 12 weeks,all rats were sacrificed and then a series of data were evaluated such as serum glucose,serum creatinine,blood urine nitrogen,the ratio of kidney weight to body weight,urinary albumin and urinary α-MG,β-MG.The structure of renal tissue was observed by electron microscope.The protein expressions of α-SMA,collagen I and PPARγ in kidney were examined by immunohistochemical stain.Results Serum glucose,urine albumin,α-MG,β-MG,the ratio of kidney weight to body weight,the expression of α-SMA and collagen I in kidney tissue were significantly lower(P0.05)and the expression of PPARr was higher(P0.05)in the high dose RGZ-treated group than in diabetic group without RGZ treatment.Conclusions The high dose of rosiglitazone has protective effects on diabetic nephropathy.
