Determination of Oleanolic Acid and Ursolic Acid Content in Dracocephalum moldovia by RP-HPLC

Abstract

Objective: Use the method of HPLC to determine the conten of oleanolic acid and ursolic acid in Dracocephalum moldovia.Method: The content of oleanolic acid and ursolic acid was detected by HPLC.The separation was carried out on a Kromasil C18 column(4.6 mm×250 mm,5 μm).The mobile phase was methanol-0.2% sodium dihydrogen phosphate(88∶ 12);the flow rate was 1.0 mL·min-1;the detective wavelength was set at 210 nm.The column temperature was at 25 ℃.Result: The linear ranges of oleanolic acid and ursolic acid were 0.42-2.10 μg(r=0.999 4) and 0.436-2.18 μg(r=0.999 7),respectively.The average recoveries were 98.9% with RSD of 1.14%(n=6) and 98.1% with RSD of 1.80%(n=6),respectively.Conclusion: This method was rapid,accurate and reliable;it could be used to determine the content of oleanolic acid and ursolic acid in D.moldovia simultaneously.
