Determination of Ursolic Acid and Oleanolic Acid Content in Gold Leaf Elder by HPLC

Abstract

Objective:To establish a HPLC method for the determination of oleanolic acid and ursolic acid content in the stems and leaves of gold leaf elder.Methods:Thermo ODS-2HYPERSIL(250 mm×4.6 mm,5 μm) was used.Mobile phase was -0.4% methanol phosphoric acid(80%:20%),with 210 nm of detection wavelength,0.8 mL/min of flow rate,25℃ of column temperature.Results:The linear range of the oleanolic acid was 12.17~243.36 μg/mL(r=0.999 9),with 98.50%(RSD 3.03) of recovery rate.The linear range of the ursolic acid was 3.25~64.92 μg(r=0.9998),with 99.49%(RSD 1.37) of recovery rate.The oleanolic acid content in the gold leaf elder stems(mg/g) was 1.485,and the ursolic acid was 0.348;but the oleanolic acid in the leaves was 0.636,and the ursolic acid was 0.437.Conclusion:The method is simple and accurate and can be used for the determination of the oleanolic acid and ursolic acid content in the gold leaf elder.
