Abstract
Using bulblet of Lilium orential manissa section as the explants,its propagation tissue culture techniques of stem tips were studied.The results revealed that survival rate of explants reached 72.0% by the surface sterilization with 6% NaClO solution for 10 minute and 0.1% HgCl2 solution for 10 minute;Cut stem tip cultured in the MS+KT 0.4 mg/L+2,4-D 1.0 mg/L,callus induction frequency can reach 88.3%.The concentration ratio between cytokinin and auxin in tissue culture can effectively control the process of buds and roots occurred on plant tissue culture.In this study,the best medium formula for adventitious bud induction was the MS+ KT 1.0 mg/L+IBA 0.5 mg/L,differentiation factor of bud seedling up to 3.44,and 1/2 MS+IBA0.4 mg/L+ NAA0.3 mg/L was suitable for its rooting,rooting rate reach 96.7%.The seedlings transplanted after 40 d of nurturing,survival rate reached above 96%.
