Abstract
Objective:To study the influence of aloe-emodin on Fas ligand mRNA expression of human colon cancer cells line SW620 and invasive ability with cell culture.Methods:The inhibition concentration 50%(IC_50) of aloe-emodin was detected by MTT assay.Different concentrations(0.5IC_50,IC_50) of aloe-emodin were used in the cultured human colon cancer cells line SW620.The changes of Fas ligand mRNA were detected by the reverse transcription polymerase chain reaction method(RT-PCR)and invasive ability of SW620 cells was measured by transwell-ECM.Results:RT-PCR method revealed that the higher concentration of aloe-emodin upregulated Fas ligand mRNA expression on SW620 cells;Transwell-ECM showed that the higher concentration of aloe-emodin increased SW620 cell invasion significantly.The comparisons with different concentrations of aloe-emodin had statistical significance.Conclusion:Aloe-emodin can upregulate functional Fas ligand mRNA expression of human colon cancer cells line SW620 in certain time.At the same time,the upregulative action can raise invasive ability of cancer cells.
