Abstract

Objective:To study the influence of Aloe-emodin on Fas Ligand mRNA expression of human Bladder Cancer Cell line T24 and invasive ability with cell culture. Methods:The inhibition concentration 50%(IC50) of Aloe-emodin was detected with MTT assay. Different concentrations(0.5IC50,IC50) of Aloe-emodin was used in the cultured human Bladder Cancer Cell line T24. The changes of Fas Ligand mRNA were detected by the reverse transcription polymerase chain reaction method(RT-PCR) and invasive ability of T24 cells was measured by Transwell-ECM. Results:RT-PCR method revealed that the higher concentration Aloe-emodin upregulated Fas Ligand mRNA expression on T24 cells(p 0.05) ;Transwell-ECM showed that the higher concentration Aloe-emodin increased apoptosis rate of lymphocyte induced by T24 cells(p0.05) . The comparisons with different concentrations teams of Aloe-emodin have statistical significance(p0.05) . Conclusion:Aloe-emodin can downregulate functional Fas Ligand mRNA expression of human bladder cancer cell line T24,which presents the dosage dependence and reduces the invasiveness of bladder cancer cell.
