Abstract

OBJECTIVE To determine three effective constituents of curcumenol,curzerenone and germacrone in Rhizoma Curcumae from different areas. METHODS A HPLC method was established using a ODS-C18 column (4.6 mm×250 mm,5 μm).The mobile phase by a gradient program was methanol-water (0.4% acetic acid)-isopropanol. The flow rate was 1.0 mL·min-1 with UV detection at 254 nm. The column temperature was 35 ℃. RESULTS The method displayed good linearity of curcumenol,curzerenone and germacrone within the concentration ranges of 5.18-155.5,5.902-177.1 and 6.19-185.7 mg·L-1. The average recovery was 99.6%,101.9% and 99.8% for curcumenol,curzerenone and germacrone respectively. The RSDs the three constituents from Curcuma phaeocaulis Valeton in Sichuan Xiansen company was 1.31%,0.94% and 2.18% respectively (n=3). CONCLUSION This method is convenient,accurate and can be used to identify and evaluate the quality of Rhizoma Curcumae.
