Effects of Cultural Conditions on Flavonoids Formation in Different Explant Callus of Glycyrrhiza uralensis Fisch

Abstract

[Objective] The research aimed to screen the best explant,medium formula and cultural conditions for producing flavonoids of secondary metabolites.[Method] Under different cultural conditions,the effect of 5 exogenous hormones of ZT,ABA,NAA,KT and 6-BA on formation of flavonoids in callus induced by roots and buds of Glycyrrhiza uralensis Fisch was studied by using the methods of orthogonal experiment design of L16(45) and contrast experiment design.[Result] The T-test indicated that weak light(100-300 lx) was beneficial to forming of flavonoids production;the callus induced from root was a suitable explant for forming of flavonoids yield.F-test showed that 5 exogenous hormones had extremely significant difference on forming of flavonoids production,the effect on total flavonoids content in the callus induced by root was in the such order as: KT6-BAZTABANAA;the effect on total flavonoids content in the callus induced by bud was in the such sequence as: 6-BAKTNAAABAZT.The optimal hormone combinations obtained after optimization were: ABA 1.0 mg/L + KT 2.0 mg/L +ZT 1.0 mg/L +6-BA1.0 mg/L + NAA 2.0 mg/L.[Conclusion] The study provided a theoretical basis for industrialized production of secondary metabolites of flavonoids.
