Influence of culture conditions on the growth of callus and content of total flavonoids in Acer buergerianum Miq.

Abstract

The objective of this research was to use MS medium to analyze the influence of light,temperature and plant growth regulators on the growth of callus and content of total flavonoids in Acer buergerianum Miq. Results showed that light had an extremely significant influence on the accumulation of total flavonoids,and a significant influence on the growth of fresh weight of callus,whereas its influence on the growth of dry weight of callus was not significant. The optimum temperature for the growth of callus and accumulation of total flavonoids was 25℃. The growth of callus and accumulation of total flavonoids were suppressed both at low-temperature(15℃) and high-temperature(35℃). Compared with the constant temperature at 25℃,temperature shifts(15℃ to 25℃ and 25℃ to 35℃) reduced the growth of callus as well as the accumulation of total flavonoids. Effects of four kinds of plant growth regulators on the growth of leaf callus and accumulation of total flavonoids were extremely significant. The optimum combination of plant growth regulators for the growth of leaf callus was 0 mg/L 2,4-D+1.0 mg/L 6-BA+ 0.5 mg/L NAA+0.5 mg/L IBA,while the optimum combination for the accumulation of total flavonoids was 0 mg/L 2,4-D+1.5 mg/L 6-BA+1.0 mg/L NAA+1.0 mg/L IBA.
