Abstract
The tissue culture responses of mature seeds and radicel were compared.Callus induction were studied in four cultivars of Poa pratensis L.Effect of 6-BA concentration,callus_age and other factors on differentiation ability were studied.Results showed that mature seed as explants had higher appearance rate than radicel;MS medium,supplemented with 2mg/L 2,4-D and 0.1mg/L BA was the best medium for cultivar'Mardona'of Poa pratensis L.,callus induction rate was 58.3%;MS medium,supplemented with 3mg/L BA and 0.2 mg/L KT was suitable for differentiation medium,regeneration rate was 70%; with subculture times increased,it still keep differentiation ability.Selecting compact,friable and grown rapidly callus subculture,can always provided good plant material for gene transfer for long term.
