Effect of Kuntongning capsule on experimental dysmenorrhea and its analgesic mechanism

Abstract

Objective To explore the effects and the mechanism of Kuntongning capsule(KTNC) on primary dysmenorrhea.Methods(1) Effect of KTNC on smooth muscle contract of isolated uterus in normal rat was investigated.40 SD rats were randomly divided into five groups.After rat uterus was isolated,uterus in solvent group was treated by solvent control.Uterus in Tongjingbao group was treated by Tongjingbao,50μg/mL.Uterus in KTNC low dosage group was treated by low dosage KTNC(25 μg/mL).Uterus in KTNC medium dosage group was treated by medium dosage KTNC(50 μg/mL).Uterus in KTNC high dosage group was treated by high dosage KTNC(100μg/mL).Changes of uterus contract after treatment were observed.(2)To observe the effects of KTNC on contract of uterus smooth muscle contract caused by oxytocin in vitro.Medication was same as(1).Changes of uterus contract 20 min after treatment were observed.(3) Effect of KTNC on pain induced by acetic acid was observed;Frequency of stretching was recorded 30 min after treatment.(4) To observe the effect of KTNC on mice's swollen ears caused by xylene.After treatment degree of mice's swollen ears were observed.(5) To observe the effect of KTNC on hemorheology of normal rats.KTNC were divided into low(0.4 k/kg),medium(0.8 g/kg) and high dosages(1.6g/kg) groups.After treatment indexes of hemorrheology were observed in each group.Results(1) Compared with solvent group,the high dose groups of KTNC decreased uterus smooth muscle contract tension of normal rats significantly(P0.05).(2) KTNC groups inhibited uterus smooth muscle contract induced by oxytocin(low dose group P0.05,high dose and medium dose groups P0.01).(3) In comparison with model group,KTNC groups reduced the writhing frequency of mice in 30 min obviously(low dose group P0.05,medium,high dose group P0.01).(4) The degree of ear turgidity of mice induced by xylene in KTNC groups was lower than that in model group(P0.01).(5) The effect of KTNC on hemorheology variety of normal rats KTNC 1.6 g/kg significantly decreased whole blood viscosity and hematokrit(P0.05).Conclusion KTNC has definite effect on analgesic,antiinflammatory and improving hemorheology.
