Abstract
[Objective]To establish a rat model of fatty liver based on alcohol and high fat diet.[Methods]Female Wistar rats were randomly divided into the control group and the model group. Rats in the control group were fed by normal food and water,and given the normal saline via intragastric administration daily. Rats in the model group were feed by high fat food and 15% alcohol water,and given the 50% alcohol solution via intragastric administration daily. The experiment was lasting for 5 weeks. The levels of triglyceride( TG),total cholesterol( TC),aspartate aminotransferase( AST) and alanine aminotransferase( ALT) in serum,as well as TG and TC in hepatic tissue were measured. The pathological condition of liver was observed.[Results]The index of liver in the model group was higher than that in the control group,but the difference was not significant( P 0. 05). The serum TC level in the model group [( 2. 93 ± 0. 62) mmol / L] was higher than that in the control group [( 2. 27 ± 0. 28) mmol / L] significantly( P 0. 05). The serum TG level increased,but the difference was not significant( P 0. 05). The levels of TG and TC in hepatic tissue increased significantly( P 0. 05). There were no significant differences in serum AST and ALT activity between two groups( P 0. 05). The food utilization rate in the model group was significantly lower than that in the control group( P 0. 05). The pathological section showed that the obvious fatty degeneration was observed in hepatic tissue of rats in the model group.[Conclusion]The fatty liver model of rat is successfully established by applying the alcohol and high fat diet for 5 weeks.
