Change of Hydrogen Sulfide in Hepatic Tissue of Rats with Nonalcoholic Fatty Liver Disease

Abstract

Objective To observe the change of Hydrogen sulfide(H2S) in liver of rats with Non-alcoholic fatty liver disease(NAFLD),and to study the pathogenesis of NAFLD.Methods The male Sprague Dawley(SD) rats were randomly divided into severe group with NAFLD(severe group),mild group with NAFLD(mild group),blank control group(blank group).Severe group was given high fat diet 20 g/(Rat·day),the feed and surplus were weighed.Mild group was given high fat diet 10 g/(Rat·day),as the same weight as severe group.Blank group was given normal feed 20 g/(Rat·day).After 12 weeks,rat was killed one per week to observe the degree of Fatty Liver.The experiment ended at 16 weeks when the severe group reached the degree of severe Fatty Liver.Collected blood from abdominal aorta,obtained plasma and tested the level of triglyceride(TG) and cholesterol(TC).The homogenate of liver was used to determine the contents of H2S.Results ①TC and TG level:After feeding the TG and TC in severe group were(8.45±1.19) and(2.43±0.38) mmol/L,respectively,and in mild group were(5.89±1.24) and(1.82±0.41) mmol/L.There was a significant difference between three groups(P0.01).The TG and TC of severe group were higher than mild group,and there was a significant difference(P0.01);②The TG level in liver:TG in severe group and mild group were(0.82±0.13) mmol/L and(0.62±0.14) mmol/L,respectively,there was a significant difference compared with blank group(P0.01);③The H2S level in liver:Compared with control group,the H2S level in severe group and mild group decreased(P﹤0.01),and the H2S decreased significantly in severe group(P0.01).Conclusion The degree of fatty liver is related to TG and TC metabolic disorder,H2S participated in the NAFLD morbidity.The intervention of H2S metabolism may be the future direction of preventing and treating fatty liver disease.
