RP - HPLC Determination of Mycophenolic Acid in Human Plasma of Renal Transplantation Patients

Abstract

Objective:To develop a RP - HPLC method for the determination of the concentration of mycophenolic acid (MPA) in human plasma. Methods:The plasma sample was extracted with 3 mL chloroform (CHC13) ,then separation was achieved on the Symmetry C18 (150 mm×3. 9 mm,5μm) column. The mobile phase consisted of acetonitrile -0.05% phosphoric acid (34:66) and the flow rate was 1.0 mL·min-1. The samples were injected manually and they were measured at UV 254 nm. Naproxen was used as the internal standard. Results: The retention times for MPA and naproxen were 11. 3 min and 14. 9 min respectively. The calibration curves were linear over the range of 0. 1-40 mg·L-1. The limit of detection was 0. 1 mg ·L-1. The methodology recovery was 98. 3% -100. 4%. The extraction recovery of MPA was 88. 1% -92. 4% The intra - day RSD was 0. 98% -1.5% and inter - day RSD 1. 0% -4.4%. Conclusion:The assay was simple,rapid,and sensitive. It is satisfactory to be used on study of the pharmacokinetics and monitor the concentration of MPA in renal transplantation patients.
