Determinations of Active Compositions in Cortex Phellodendri Chinensis and Cortex Phellodendri Amurensis from Different Areas by HPLC

Abstract

Objective To determine the contents of active compositions of in different areas of Cortex Phellodendri Chinensis and Cortex Phellodendri Amurensis,to provide some academic basis for quality criterion of Cortex Phellodendri Chinensis.Methods The method of HPLC was used to determine the content of berberine hydrochloride,palmatine hydrochloride and jatrorrhizine hydrochloride in different areas of Cortex Phellodendri Chinensis and Cortex Phellodendri Amurensis.The method was established on a Kromasil C18 column with the mobile phase consisting of acetonitril-0.1% phosphonic acid(50:50)(per 100 mL plus sodium lauryl sulfate 0.1 g) and flow rate of 1.0 mL/min and the temperature at 30 ℃,the detection wavelength was 265 nm.Results The results showed that the highest content was berberine hydrochloride in Cortex Phellodendri Chinensis,between 3.73% ~ 6.131%;the content of jatrorrhizine and palmatine were low,between 0.007% ~ 0.061% and 0.027% ~ 0.082%,respectively;the lowest content was jatrorrhizine in Cortex Phellodendri Amurensis,between 0.008% ~ 0.043%,berberine hydrochloride between 0.474% ~ 1.224%,palmatine between 0.516% ~ 1.224%.Conclusion The content of active compositions was significantly different in different areas of Cortex Phellodendri Chinensis and Cortex Phellodendri Amurensis,the content of the two species was quite different.
