Determination of tetrahydropalmatine content in Corydalis yanhusuo W.T.Wang from different habitats by RP-HPLC

Abstract

The content of tetrahydropalmatine in Corydalis yanhusuo W.T.Wang from different habitats was determined by RP-HPLC method.A kromasil C_(18) column(200 mm×4.6 mm,(5 μm)) was used with the mobile phase consisting of methanol-0.1% phosphoric acid(with pH value adjusted to 6.09 with triethylamine)(V∶V=55∶45).The flow rate was 1.0 mL·min~(-1),and the detection wavelength was set at 280 nm.The linear range for detection of tetrahydropalmatine was 22.0~352.0 μg · mL~(-1)(r=0.999 9).The average recovery rate of the method was 98.6%,with RSD=0.4%(n=9).The method can be used for the control over quality of Corydalis yanhusuo W.T.Wang and for comparing the content of tetrahydropalmatine originated from different habitats.
