Abstract
Objective:To observe in vitro the influence of platelet-rich plasma(PRP) in different concentration on the activity of alkaline phosphatase from auto bone marrow stromal cells, and to evaluate the biological effect PRP induced difterentiation from bone marrow stromal cell to osteoblasts. Methods: After abstraction of blood in vein and bone marrow of dog , PRP in different concentration were prepared by modified Appel method . Sufficient stable BMSC were acquired through subculture screening technique, and the activity of alkaline phosphatase was assayed by methods of enzyme kinetics. Results: 1. Bone marrow stromal cells after subculture began to adhere after 24 hours and the cells fused to single layer after 12 days .Most of them took on the spindle-shaped conformation of fibroblast and gradually turned to rectangle or multiangle after subculture by induced solution. 2. Assay the activity of alkaline phosphatase with methods of enzyme kinetics in every group on the 3rd, 6th, 9th and 12th day respectively. After paired t-test , the activity of ALP on the 6th, 9th, 12th day was significantly higher than that on the 3rd day and the activity in lower concentration PRP group was more active than that in higher group. The difference is significant in statistics (P0.05 ) . Conclusions: PRP can promote the activity of ALP in bone marrow stromal cells and can also accelerate effectively the differentiation from BMSC to pre-mature osteobalasts .
