Study of Cell Growth Inhibition and Apoptosis Induced by Oxaliplatin in Human Lung Adenocarcinoma Cell Line-A549

Abstract

Objective To research in vitro chemosensitivity and apoptosis induced by oxaliplatin in human lung adenocarcinoma cell line-A549. Methods A549 was incubated with oxaliplatin of virous concentration , cell growth inhibition was assesed by MTT colorimetric assay and apoptotic rate was detected by FCM. Results At dose of 100,50,25,12.5 μg/ml, oxaliplation apparently inhibited growth of A549 cell, FACS can showed apoptotic rate was 35%,13.6%,2.6%,1.4% for 24 h treatment at dose of 100,50,25,12.5 μg/ml respectively. Conclusion The cell growth inhibition of oxaliplatin on A549 is dose-dependent and time-dependent, oxaliplatin can induce apoptosis in A549 cell which is dose-dependent. There is a positive linear correlation between apoptosis assessed by FACS can and growth inhibition rates by MTT( P 0.01).
