Effect of growth inhibition and apoptosis induced by oxaliplatin on human nasopharyngeal carcinoma cell line CNE-2 in vitro

Abstract

Objective:To find more effectively chemotherapeutic agents and treatment regimens,we studied the effects of oxaliplatin on cell growth inhibition and apoptosis induction in human nasopharyngeal carcinoma cell line CNE-2.Methods:CNE-2 cells were incubated with oxaliplatin at various concentration and time,cell growth inhibition was assessed by Sulphorhodamine assay,cell-cycle kinetics and apoptosis were analyzed by fluorenscence microscopy and flow cytometry.Results:The growth of CNE-2 cells was significantly inhibited by oxaliplatin in a dose-dependent and time-dependent fashion.The inhibition of cell growth was(97.46±1.76)% after incubation with 80μg/ml oxaliplatin for 48 hours.oxaliplatin could cause CNE-2 cells cycle to change.With the increase of drug concentration,G2/M phase cell percentage increased.The early-stage and late-stage apoptosis rate of CNE-2 cells were(4.48±1.23)%and(9.48±2.30)%after incubation with 80μg/ml Oxaliplatin for 48 hours respectively.Conclusions:Oxaliplatin suppresses the growth of CNE-2 cells in vitro by causing cell-cycle arrest and cell apoptosis.
