Abstract
Objectives: To investigate the effects of valsantan on the prevention and treatment of diabetic nephropathy. Methods: The effect of valsantan on the proliferation of cultured human mesangial cells was measured by MTT. RT-PCR , ELISA and RIA etc were used to measure the effect of valsantan on the expression of AM mRNA , TGF-β1 mRNA and content of AM, AngⅡ, laminin, type Ⅳ collagen, fibronectin of the cultured human mesangial cells under the high glucose condition in vitro . Results: Valsantan could effectively suppress the excessive expression of TGF-β1 mRNA and AM mRNA of mesangial cells under high glucose concentration, and they could also decrease the concentrations of TGF-β1, AM, AngⅡ, laminin and type Ⅳ collagen in the supernatants. In addition, valsantan could suppress the proliferation of cultured human mesangial cell under high glucose conditions. Conclusion: Valsantan has renal protective property that was associated with its suppression on the expression of TGF-β1 and the proliferation of human mesangial cell cultured under high glucose conditions. At the same time, it also adjusted the expression and secretion of AM.
