Abstract
Objective: To evaluate the effect of HMG-COA reductase inhibitor (Simvastatin) and angiotensin converting enzyme inhibitor (Cilazapril) on the proliferation of cultured human mesangial cells and the secretion of extracelluar matrix proteins. Methods: By using the high concentration of glucose, Simvastatin and Cilazapril to stimulate the cultured human placenta glomeruli mesangial cells in vitro, we examined the effect of different duration, different concentration, single or associated medication on the proliferation of mesangial cells and transforming growth factors-Pi (TGF-β1), fibronectin, laminin and type IV collagen in the supematants. Results: High concentration of glucose can promote the proliferation of cultured human mesangial cells, and increase the concentrations of TGF-B1, fibronectin, laminin and type IV collagen in the supematants, combinations both of Cilazpril and Simvastatin had stronger suppression on the proliferation of culture human mesangial cell and the secretion of TGF-β1, fibronectin, laminin and type IV collagen than that in each one alone. Conclusion: Simvastatin and Cilazapril have suppressive action on the proliferation of mesangial cells and the secretion of extracelluar matrix proteins under high concentration of glucose. The two drugs in combination have more suppressive action.
