Abstract

OBJECTIVE To develop a RP-HPLC method for the determination and analysis of paeonol in extract of cortex moutan. METHODS The content of Paeonol was determined by RP-HPLC method. The analysis was performed on a RP-HPLC apparatus equipped with a Inertsil ODS-3 C 18 column( 4. 6mm × 250mm,5μm),The DAD detection was carried out at the wavelength of 274nm and a temperature of 30℃,the mobile phase consisted of methanol-water( 60∶ 40) with the flow rate of 0. 8mL·min- 1. RESULTS The calibration curve of paeonol was linear in the ranges of 35. 8μg·mL- 1( R2= 0. 9999). The average recovery of paeonol was 98. 54%( RSD = 1. 03%,n = 6). The sample was extracted with water-steam distillation after ultrasonication 30min. CONCLUSION The method is effective,accurate,and can be applied to quantitatively analyse paeonol in extract of cortex moutan.
