Abstract
AIM To study the effect of TNF α on apoptosis of vascular endothelial cells(VEC) in early second thrombosis after skin avulsion. METHODS It was divided into experimental group (EC+TNF α) and inhibitory group (rhTNF αAb pretreatment of EC+TNF α). According to various times and concentration, apoptosis rate of vascular endothelial cells was measured by FCM for experimental group and inhibitory group. RESULTS TNF α caused massive death of endothelial cells in primary culture in a concentration and time dependent manner. Apoptosis rate of EC was (2.6±0.42)% ~(40.5±8.3)% from 0 to 48 h and (2.6±0.42)% ~(13.7±2.6)% from 0 to 3000 u·mL -1 in experimental group,and it de creased significantly in inhibitory group. CONCLUSION TNF α causes apoptosis of EC in a concentration and time dependent manner and rhTNF αmonoclonal antibody can decrease cytotoxic for EC caused by TNF α.
