Abstract
Objective:To investigate the effect of NiaoSuanLiXian containing serum on expression of vascular cell adhesion molecule(VCAM-1) mRNA in vascular endothelial cells induced by TNF-α. Methods:Vascular endothelial cells was cultured and stimulated with different density(12.5,25,and 50 μg/ml) of TNF-α in vitro,.At the same time,the cells were co-incubated with 25 μg/ml density of TNF-α for 8,16,24,and 48 hours.They were interved by different density(2.5%,5%,and 10%)of NiaoSuanLiXian containing serum.The expression of VCAM-1 mRNA in vascular endothelial cells was detected by RT-PCR. Results:The expression of VCAM-1 mRNA in vascular endothelial cells induced by TNF-α increased significantly(P0.01),but no difference was noted between different density groups.The expression of VCAM-1 mRNA in vascular endothelial cells induced by the 25 μg/ml density of TNF-α increased significantly after 16,24,and 48 hour incubation(P0.01).NiaoSuanLiXian containing serum at all experimental density significantly down-regulated the expression of VCAM-1 mRNA(P0.05,P0.01). Conclusion:Down-regulating the expression of VCAM-1 in vascular endothelial cells may be one of the mechanisms of NiaoSuanLiXian reducing the inflammatory injures.
