IGF-1 protection from cardiomyocytes apoptosis induced by TNF-αvia a PI3K/Akt-dependent pathway

Abstract

[Objective] To investigate the effects of insulin like growth factor 1(IGF-1) on apoptosis of cardiomyocytes induced by tumor necrosis factor-α(TNF-α) and its possible relationship with PI3K/Akt.[Methods] Rat cardiomyocyte isolated according to collagenage perfusion protocol.Cardiomyocyte primarily cultured for 1 days,were separated into four groups: control group,TNF-α group,IGF-1+TNF-α group and IGF-1+TNF-α+LY294002 group.Cells morphological transformation was observed under microscope.Cardiomyocyte apoptosis was determined by MTT.[Results] The number of the apoptotic cardiomyocyte in TNF-α group was significantly lower than that in control group.The number in IGF-1+TNF-α group was significantly higher than that in TNF-α group,and had no significant differences when compared with the control group.Apoptotic cardiomyocyte in IGF-1+TNF-α+LY294002 group were significantly lower than those in IGF-1+TNF-α group.The differences was significant between IGF-1+TNF-α+LY294002 and TNF-α group.[Conclusions] IGF1 may protect the cardiomyocyte apoptosis induced by TNF-α,and PI3K/Akt pathway may play an important role in this protection.
