Comparison of Different Methods of DNA Extraction of Peanut Leave

Abstract

Objective:The objective of the paper is to select the best method to extract DNA from peanut leave.Methods: 4 methods were investigated,including SDS,proved SDS,CTAB and proved CTAB extract approach.The electrophoresis results showed that DNA extracted by SDS approach degrades seriously,that by the improved SDS approach with too much glucide,and that by Improved CTAB approach trailed slightly.Results: The results also showed that CTAB approach was better than improved CTAB approach.The extract ratio of two SDS methods were higher than that of the two CTAB methods while the OD value of two SDS methods were lower than that of the two CTAB methods(ODCTAB≈1.80).Conclusion: Compared with CTAB,the improved CTAB approach was more simple,time saving and with higher extract rate.Considered above all,we took Conclusion that CTAB was the best method to extract peanut leaf DNA.
