Abstract
Objective:To investigate the regulating effect of ulinastatin on the serumal level of TNF-α,IL-6 and IL-10 with sepsis in the rats.Methods:Male SD rats were randomly divided into four groups:normal control group,sham group,cecal ligation and puncture(CLP) group and ulinastatin pretreated group.Cecal ligation and puncture(CLP) were performed to make sepsis model.Rats were sacrificed at 2.0,8.0,12.0 and 24.0 h,the level of the TNF-α,IL-6 and IL-10 were examined with ELSA and left lung tissue was obtained to observe the pathological changes under microscope.Results:In comparison with the sham group,the level of the TNF-α,IL-6 and IL-10 increased in CLP group(P 0.05).The level of TNF-αin ulinastatin pretreated group was much lower than that in CLP group(P 0.01) besides 24.0 h group.The level of IL-6 in ulinastatin pretreated group was much lower than that in CLP group(P 0.01).The level of IL-10 in the CLP group showed significant difference from the ulinastatin pretreated group(P 0.01).HE-stained sections revealed that ulinastatin attenuated lung injury.Conclusion:The ulinastatin can downregulate the level of the TNF-αand IL-6 can upregulate the level of the IL-10 obviously and attenuate lung injury in sepsis.
