Simultaneous Determination of Two Crocin from Saffron in Zhenlongxingnao Jiaonang by HPLC

Abstract

Objective A HPLC method was established for the determination of Crocin Ⅰand Crocin Ⅱ from Saffron in Zhenlongxingnao Jiaonang. Methods The analysis was carried out on a Waters C18 column(250 mm×4.6 mm, 5 μm) with mobile phase of Acetonitrile-1% acetic acid aqueous solution(25:75). The flow rate was 1.0 ml/min; the UV detection wavelength was set at 440 nm, and the column temperature was room temperature. Results There was a good linear relationship within the range of 6.03μg/ ml~60.30μg/ml, 3.01μg/ml~30.10μg/ml for CrocinⅠ, CrocinⅡ, respectively. The correlation coefficient r was separately 0.9997 and 0.9999. The average recovery of Crocin Ⅰ, Crocin Ⅱ was 97.80%, 97.44%, respectively. And the RSD was 0.66%(n =6), 1.71%(n=6), respectively. Conclusion The established method can determine CrocinⅠ, CrocinⅡ simultaneously. It can be used for the quality control of Zhenlongxingnao Jiaonang.
