Abstract
Taking leaf of Chrysanthemum morifolium ‘Qiandaiji'as material,tissue culture conditions and media for callus induction,cluster buds propagation and rooting were studied in this article.The results showed that the proper medium of callus induction was SH+0.5mg/L 6-BA+1.0mg/L NAA and resulted in an induction efficiency of 100%.The cluster buds well grow in MS+2.0mg/L 6-BA+0.2mg/L NAA.And the appropriate rooting medium was 1/2MS+0.1mg/L NAA.All three types of media were contained 30g/L sucrose and 6.0g/L agar with pH 5.8.The culture conditions were optimized to 2 500 lx light intensity,25℃temperature and 12hlight/12 hdark circle.
