Determination of oleanolic acid and ursolic acid in Eriobotrya japonica(Thumb.)Lindl extractive by HPLC

Abstract

OBJECTIVE To establish a method for determination of oleanolic acid and ursolic acid in Eriobotrya japonica (Thumb.)Lindl extractive.METHODS HPLC method was used.The samples were separated on a C 18 (250 mm× 4.6 mm , 5 μm )column with the mobile phase of methanol water(94∶6),which contained 0.08% lacial acetic acid.The detection wavelength was set at 220 nm .Flow rate was 0.6 mL·min -1 .Column temperature was 25 ℃ .RESULTS The linear ranges of determination of oleanolic acid and ursolic acid were 0.45 - 2.70 μg and 0.46 - 2.76 μg respectively.The average recoveris of 98.3% with RSD of 3.15% ,for oleanolic acid and 97.7% with RSD of 2.97% for ursolic acid were obtained.CONCLUSION The method is sensitive,rapid and accurate and suitable for the determination of oleanolic acid and ursolic acid in Eriobotrya japonica (Thumb.)Lindl extractive.
