Abstract
Objective: To delve into the expression and clinical significance of the protein of the suppressor gene P16, and oncogene cyclinD1 in human endometrial carcinoma, and to find out the correlation between their expression and clinical stages, pathological grades and the degree of myometrial invasion of endometrial carcinoma. Methods: ABC immunohistochemical assay was used to observe the of expression P16 and cyclinD1 protein in 41 cases of endometrial carcinoma, 15 cases of complex hyperplasia endometrium, and 8 cases of normal endometrium. The differences of expression related to different histological grades, degrees of myometrial invasion and clinical stages of endometrial carcinoma were observed. Results: The positive rate of P16 protein expression in endometrial carcinoma was 51.2%, significantly lower than that in normal endometrium(P0.01) and complex hyperplasia endometrium(P0.05). The positive rate of cyclinD1 expression in endometrial carcinoma was 56.1%, which was significantly higher than that in normal endometrium(P0.01) and complicated hyperplasia endometrium(P0.01). The expression of P16 gene diminished with higher pathological grades, greater degrees of myometrial invasion and advaneed clinical stages, but the expression of cyclinD1 enhanced. Synchronistic abnormal expression of the two parameters usually correlated with endometrial carcinoma of grade Ⅲ, with deep invasion of myometrium and at stages Ⅲ or Ⅳ. Conclusion: Abnormal expression of P16 gene and cyclinD1 were closely related to occurrence and development of the endometrial carcinoma. They may be ueed as parameters to direct clinical therapy and to evaluate the prognosis of endometrial carcinoma.
