Abstract
To establish HPLC method for the determination of fermentation liquid ammonia acid tryptophan and tyrosine. Agilent C18 column(250mm×4.6mm,5μm)was used. The mobile phase was 0.1mol/L sodium acetate(pH 4.0±0.05)-Methanol(90∶10). The flow rate was 1.0mL·min-1and detection wavelength was 280nm. Column temperature was 30℃. The results showed that tryptophan linear between 50~500μg·mL-1. The correlation coefficients was r=0.9985. The average recoveries was 99.3%(n=4). Tyrosine was linear between 20~2000μg·mL-1. The correlation coefficients was r=0.9991. The average recoveries was 100.2%(n =4). So drew the conclusions:Direct determination method without the need of derivative is simple,accurate, for the determination of content of fermentation liquid tryptophan and tyrosine.
