Abstract
IM To understand the relationship between the expression of both ras and p53 gene RNA and protein and the metastasis of primary hepatocellular carcinoma (HCC).METHODS Immunohistochemistry and slot blot were used to study ras and p53 gene protein and RNA in 14 cases of HCC with metastasis and 16 cases of HCC without metastasis respectively.RESULTS After microwave treatment, the positive rates of both p21 and p53 protein in 14 cases of HCC with metastasis were significantly higher than that without metastasis (11/14 vs 5/16, P=0014 and 12/14 vs 7/16, P=0026, respectively). The results of slot blot showed that OD value of Nras RNA in HCC with metastasis was 3 to 5 times higher than that in HCC without metastasis, whereas OD value of p53 RNA had no difference between the two groups.CONCLUSION Immunohistochemistry of p21 and p53 protein after antigen retrieval is a useful index for patients with HCC metastatic potential
