Abstract
Objective:To establish an RP-HPLC method for determination of chlorogenic acid in Flos Lonicerae Hypoglaucae from Guangxi.Method:Hypersil ODS-2(4.6 mm×250 mm,5 μm) was adopted; The mobile phase was methanol-acetonitrile-0.05% phosphoric acid with gradient elution, and the flow rate was 0.5 mL·min-1;The detective wavelength was 238 nm and column temperature was 30 ℃.Results:Chlorogenic acid was determined on 23 samples of Flos Lonicerae Hypoglaucae from different producing areas and processing methods. The linear range of chlorogenic acid was 0.2-1.0 μg(r=0.9999);The average recovery(n=6) was 100.5%(RSD=2.8%).Conclusion:The difference between the content of chlorogenic acid in different samples with different origins, different processing methods was marked. The content of chlorogenic acid in all samples is far higher than the standard of Chinese Pharmacopoeia.
