Callus induction and differentiation with diverse explants of Acacia mearnsii

Abstract

To establish an efficient tissue culture system for rapid propagation of Acacia mearnsii,the leaves,stem segments and hypocotyls were used as explants and cultured on MS medium. And effects of different types and concentrations of plant growth regulators on the induction and differentiation of callus were examined. The results showed that for the induction rate and the growth of callus,leaves were the best of three explants with the suitable medium of MS + 1. 5 mg / L 6-BA + 0. 005 mg /L TDZ + 1. 0 mg / L IBA. The callus of leaves and hypocotyls could induce adventitious buds. Moreover,different explants had a significant effect on differentiation rate. As for the adventitious buds differentiated from leaves' callus,the best differentiation value up to 32. 60% was gained on medium of MS + 0. 01 mg / L TDZ + 0. 2 mg / L IBA.
