Abstract

[Objective]The present experiment was conducted to study the phylogenetic relationship amongst Guangxi mulberry(Morus atropurpurea Roxb.)germplasm resources using SRAP(sequence-related amplified polymorphism) markers.[Method]The polymorphism amongst 28 Guangxi local mulberry germplasm resources,belonging to Guangdong mulberry family,was assessed using SRAP molecular marker technique and their genetic diversity was analyzed using cluster analysis.[Result]The results showed the amplification of 144 bands in 28 mulberry germplasm using 22 pairs of SRAP primer combinations.Of which,45 bands(31.81%) showed polymorphism.On an average,the number of total and polymorphic bands/primer pair primer combination was recorded as 6.55 and 2.05,respectively.The genetic similarity coefficients among mulberry resources ranged from 0.8264 to 1.0000,with an average of 0.8944.UPGMA results showed 3 clear clusters for the 28 mulberry materials in dendrogram.The first group included 20 local mulberry materials,the second included 7 self-breeding mulberry materials and the third had only one material.[Conclusion]High genetic variations amongst Guangxi mulberry germplasm resources were observed,and the genetic diversity and phylogenetic relationship of local mulberry germplasm were found in accordance with their habitat.The results of our experiment will be helpful in classification and breeding of the mulberry genotypes.
