An Analysis on Genetic Diversity of Wild Mulberry Germplasm Resources in Heilongjiang Province

Abstract

In order to facilitate an accurate identification and evaluation to the preservation and practical value of wild mulberry germplasm resources on their cold tolerance and drought resistance,genetic diversity and phylogenetic relationship among 24 wild mulberry germplasm resources collected from Heilongjiang Province was analyzed by random amplified polymorphic DNA(RAPD) molecular marker technology.From 20 RAPD random primers,4 effective primers were obtained.Amplification to genomic DNAs of these 24 wild mulberry germplasm materials yielded 275 bands,among which 273 were polymorphic.The polymorphic rate reach 99.3%.And the number of obtained bands from amplifying single mulberry germplasm material with single primer ranged from 2 to 5.Genetic distance between the 24 wild mulberry germplasm materials was within the range of 0.047 to 1.000.Among them,the highest genetic resemblance was between Dumeng 2 and Dumeng 3,and between Ning'an 1 and Ning'an 3.And the lowest genetic resemblance was between Dumeng 3 and Ning'an 3.The genetic relationship among 24 wild mulberry germplasm materials was basically in agreement with their regional distribution.
