Analysis of Genetic Diversity of 93 Mulberry Germplasm Resources of Morus atropurpurea Roxb. Using ISSR Markers

Abstract

In this study,the genetic diversity of mulberry germplasm resources of Morus atropurpurea Roxb.was investigated by means of ISSR molecular marking technology,which will provide fundamental references for the conservation and identification of mulberry germplasm resources,and the breeding of new varieties.Totally 128 bands were amplified from genomic DNA of 93 germplasm materials of Morus atropurpurea Roxb.using 13 ISSR primers and there were 94 polymorphic bands with a percentage of polymorphic band of 73.44%,implying that there was rich genetic diversity among the tested germplasm materials of Morus atropurpurea Roxb..Genetic similarity coefficients between the 93 germplasm materials of Morus atropurpurea Roxb.ranged from 0.585 9 to 0.937 5,with an average of 0.761 7.The genetic relationship between these germplasm materials was analyzed with UPGMA method based on their genetic similarity coefficients.The result showed that the 93 germplasm materials could be clustered into 6 groups at genetic similarity coefficient of 0.664,in which groups Ⅲ,Ⅳ and Ⅵ could be further divided into 2 subgroups respectively at genetic similarity coefficient of 0.685.The obtained results demonstrated that the genetic relationship of the tested germplasm materials of Morus atropurpurea Roxb.is correlated to geographical distribution and sex of flower of each germplasm material.
