Abstract

A micropropagation system was established in stem of Populus deltoides L.'35/66'.Tender stems of 35 poplar in field were been easy to sterilize in April.Axiliary buds from the stems were sprouted in WPM basal medium with 0.1 mg/L NAA,0.5 mg/L 6-BA and 0.5 mg/L GA3.Axillary buds were proliferated in WPM basal medium supplemented with 0.2 mg/L 6-BA,0.1 mg/L IAA and 0.1 mg/L IBA.Plantlets were rooted better in 3/5WPM basal medium with 0.1 mg/L IBA.Adventitious shoots were regenerated from leaf explants of rooted plantlets.The highest regeneration rate(92.86%) and clustered adventitious buds(10)was obtained in WPM basal medium with 0.2 mg/L 6-BA,0.1 mg/L IAA and 0.1 mg/L IBA.Plantlets from adventitious buds were rooted in root medium with 0.1 mg/L IBA and transplanted to the soil successfully.
