Micropropagation and Acclimatization of Pluots ("Prunus Simonfi") with Semi-Lignified Branches

Abstract

The young and semi-lignified branches with one lateral bud from Pluots ("Prunus simonfi" Cart) were used as explant to develop a regeneration protocol. The disinfection in 0.1% HgCl_2 for 8min was the optimum treatment and the optimum basal medium was Woody Plant Medium (WPM), which supplemented with 0.1 mg/L IBA + 1.0 mg/L BA + 10.0 mg/L Vitamin C (VC) + 1.0 mg/L activated carbon (AC) for adventitious shoots induction was the best, better than Murashige and Skoog (MS) medium. Adventitious shoots were established as proliferating shoot cultures following transfer to WPM medium supplemented with 0.2 mg/L IBA + 1.5 mg/L BA + 15.0 mg/L VC. The best rooting rate (100%), mean number (6.7) of roots and mean root length (19.04 cm) of in vitro shoots were obtained and the optimum medium for adventitious shoots rooting was WPM + IBA 1.0 mg/L+ BA 0.2 mg/L + AC 0.5 g/L. Rooted plantlets were successfully acclimatized to the greenhouse on medium containing humus soil + perlite + sand (1:1:1), which was the optimum medium for acclimatization (70% survival rate).
