Generation and characterization of monoclonal antibodies against canine distemper virus.

Abstract

Canine distemper virus was purified and used to immunize Balb/C mice.The hybridoma culture supernants were screened by indirect ELISA.The positive cells were subcloned until all clones were positive.The specificity of monoclonal antibodies was confirmed by Vero cells cross-reaction test and lFA.The Results showed that six hybridoma cells secreting anti-CDV McAb were obtained.These six cells were named AD7,AE2,CE3,EH5,GG6 and GF1.All the six McAbs confirmed by cross-reaction test and lFA were specificity against CDV.The positive hybridomas were also able to secrete McAbs stably by series passage in vitro with freezing and recovering.The titers of cell culture supernant and ascites tested by ELISA were 26—211 and 104—107,respectively.Additivity ELISA results showed that epitopes recognized by 4 McAbs(AE2,CE3,GG6 and GF1) differed from these by McAb AD7 and EH5.
