Protective Effects of Acetylpuerarin on Focal Brain Ischemia-Reperfusion Injury in Rats

Abstract

OBJECTIVE To investigate the protective effect of acetylpuerarin(compound N-2211)on cerebral ischemia-reperfusion injury in rats and its mechanism.METHODS Brain ischemia/reperfusion injury model was prepared by MCAO method.The activity of superoxide dismutase(SOD),the level of malondialdehyde(MDA) and T-AOC were assayed with different methods respectively.RESULTS Compared with IR-group,the level of MDA and infarction size decreased,the activities of SOD and total antioxidative capacity(T-AOC)increased in acetylpuerarin treated rats.And the neurological deficit score was significant decreased in 1 h occlusion and 24 h middle cerebral artery reperfusion rats while sham operation rats had no residual deficit(P0.01).Infarcted area was improved by acetylpuerarin in both cerebral cortex and basal ganglia in operation group stained by triphenyltetrazolium chloride staining method.CONCLUSION Acetylpuerarin possesses neural protection in brain ischemia/reperfusion damage.The positive effect of acetylpuerarin on brain focal ischemia injury may partly depend on its antioxidant property.
