Abstract

[Objective]The present experiment was aimed to study the effects of different hormone combinations on callus induction in anthers taken from tomato thermo-sensitive genic male sterile lines(TSGMSLs) and their hybrid combinations.[Method]Tomato TSGMSLs,viz.,Da 107 and Di D4,and their hybrid combinations,viz.,Da 107×Niufanqie and Di D4×Hongkunlun were used in the studies.Combinations of different hormones(CPPU,TDZ,KT) in basic MS medium(containing 1.0 mg/L 2,4-D+1.0 mg/L 6-BA) were used for anther callus induction.[Result]Adding different CPPU,TDZ or KT concentrations in basic MS medium significantly improved the induction of anther callus,and the suitable concentration of CPPU,TDZ and KT was 0.4-0.6,0.03 and 1.5 mg/L,respectively.Under the same hormone combination conditions,the anthers taken from tomato TSGMSLs showed better callus induction compared to those taken from hybrid combinations,the order of the average rate of callus induction and average diameter of callus was:Di D4Da 107Di D4×HongkunlunDa 107×Niufanqie.[Conclusion]Anthers of different tomato genotypes differed in their sensitivities to different hormones.Furthermore,different combinations of hormones also differed in affecting induction of tomato anther callus,out of them,the combinations of KT and 2,4-D+6-BA showed best results.
