Abstract
To establish an efficient regeneration system of hybrid F1of Indicaand Japonicavarieties,anthers of F1from crosses of Indicaand Japonicavarieties‘Zaxima'/‘Nanjing 46'were used as the explants to induce the callus in different low-temperature pretreatment time,different basic media,different media supplemented with different mass concentrations and combinations of sucrose and plant hormones.The result showed anthers pretreated at 4 ℃for 2-5days were induced to callus in a higher level,significantly higher than those under other pretreatments.Medium N6and M8were better than the SK3medium in callus induction.Callus induction frequency was significantly different,and the incorporation of sucrose in the mass concentration of 60g·L-1 in the callus induction medium.The 32.14%of callus inducting rate in the M8medium added with 2,4-D 2mg·L-1+ KT0.3mg·L-1 was obtained and the 50% of callus differentiation rate in the MS medium added with KT 2mg·L-1+ABA 2mg·L-1 was obtained,but the frequency of green plantlet was almost4.55%.
