Settled in transplant liver of bone marrow mesenchymal stem cells

Abstract

Objective To study the bone marrow mesenchymal stem cells settled in transplant liver.Methods Rat bone marrow mesenchymal stem cells (BMSCs) were isolated and cultured by density gradient centrifugation.The surface antigens (CD45,CD44,CD90) of BMSCs were identified by flow cytometry,the dosage and timing of 4',6-diamidino-2-phenylindole (DAPI) labeling were respectively 0.5,1.5,2.5 mg/L and 12 hours,labeling rate were observed under fluorescence microscope.Choose the optimal dosage of labeling BMSCs inject in transplant liver before portal vein cased,and cut out liver tissue in 7 days,12days for frozen pathology,observed the BMSCs settled situation under fluorescence microscope.Results Density gradient centrifugation can isolates and culture BMSCs successfully.Flows cytometry shows that BMSCs expressed CD90,CD44,CD45 is 97.1%,87.4%,1.1%.The three dosage of DAPI showed can labeling BMSCs,which the best labeling dosage is 1.5 mg/L for 95%.Fluorescence can observed in transplanted liver pathology post operation 7 days and 12 days.Conclusion BMSCs can settled in transplanted liver.
