Abstract
Objective To investigate the reversal effect tumor necrosis factor-α on cisplatin (DDP) resistance of human lung cancer cell lines A549/DDP and to explore the mechanism of reversing cisplatin resistance of A549/DDP.Methods The cytotoxic effects of tumor necrosis factor-α 250,1000 U/ml and cisplatin on A549/DDP were measured by methyl thiazol tetrazolium (MTT) assay,the apoptotic cells of 250,1000 U/ml TNF-α and cisplatin on A549/DDP were detected by flow cytometry and Annexin V-FITC/proliferation index (PI) double staining.Results The IC50 to cisplatin were decreased from 7.12 mg/L to 5.02 mg/L,4.28 mg/L respectively,and the resistance to cisplatin was reversed by 250 U/ml,1000 U/ml TNF-α visibly in A549/DDP (P <0.01 ),the reversal folds were 1.418 and 1.663 respectively.The apoptosis rate of A549/DDP increased significantly in 250U/ml,1000U/ml TNF-α plus cisplatin group than that of no-drug group and cisplatin group (P < 0.01 ).Conclusion TNF-α can reverse the resistance to cisplatin in A549/DDP cell lines,the mechanism of reversing resistance to cisplatin is probably related to inducing the apoptosis in A549/DDP.
